1) Clinical Pharmacology and Toxiology, University Hospital, Lausanne, Switzerland and sanofi-Aventis, Monipellier, france tested on the drug effect to creatine on renal. They aim to determine the effects of dronedarone on renal function and tubular cation handling. It is said that creatinine transport across tubular epithelial cell were disturbed by the drug. This make the cations compete for the pathway.


  2) The researchers select a dozen of random male as subject. Three types of drugs were used, dronedarone, inutest vials which used to determined sinistrin clearance and sodium pra-amino-hippurate vials. All the subject follow the procedure which each subject must received twice daily repeated doses of dronedarone or placebo. Samples such as urine and blood were taken to evaluate the test result. There were several method used in analytical method, high-performance liquid chromatography method used to determined plasma, urine concentration of sinistrin, PAH, and NMN, while standard Jaffe colorimetric method used to measured creatinine. Based on the formula given, CLR=(U x V)/(P x t) it used to determined creatinine, sinistrin, PAH and NMN renal clearance in the data analysis and systemic clearance contained more inertia than renal clearance represent sudden and fast changes in renal function. Last method used was statistical analysis, it performed using a linear mixed effect model with fixed terms and a term for subjects within sequence.


  3) Subject, renal function test and safety result shows compared with placebo, researcher found that, dronedarone lower the renal creatinine CL and NMN CL, but never influenced renal sinistrin CL, PAH CL and other renal parameters.


  4) The researchers indicated that dronedarone has no influence on renal clearance of sinistrin and never effect the GFR. Dronedarone decreased renal clearance by about 18% due to disturbance of the cation transport system by this antiarrhythmic drug in a way similar to cimetidine. A similar decreased observed in  the renal clearance of another cation, NMN. Creatinine clearance damage by dronedarone, though compatible with an inhibition of the OCT system, was lower than expected.


  5) I learned that this journal plays an important role in explanation of dronedarone effects in renal function. This will help the educators, research group, teaching, and physicians to understand the causes and the method use to determined problem.

Aw, this was a really nice post. In idea I would like to put in writing like this additionally - taking time and actual effort to make a very good article… but what can I say… I procrastinate alot and by no means seem to get something done.
